Patlıcanda şeker, bal ve büyüme düzenleyicilerin anter kültüründe embriyoid oluşumuna etkisi

Abstract

Anter kültüründe besin ortamı bileşenlerinden şeker ve büyüme düzenleyiciler önemli role sahiptir. Genelde besin ortamlarında karbonhidrat kaynağı olarak sakkaroz kullanılmaktadır. Anter kültürü ile ilgili birçok çalışmada sakkarozun %2-4 dozları önerilirken, az sayıda çalışmada şekerin %9-15 arasındaki dozları önerilmektedir. Anter kültürü çalışmalarında karbonhidrat kaynağı olarak şeker yerine bal kullanılması ile ilgili çok az araştırma yapılmıştır. Patlıcanda anter kültüründe şeker yerine bal kullanılmasının etkisinin araştırıldığı bu çalışmada, farklı sakkaroz ve bal dozları (30, 60, 90, 120 ve 150 mg/l) karşılaştırılmıştır. Büyüme düzenleyici olarak 2, 4-dichlorophenoxyacetic acid (2,4-D) ve Kinetinin (Kin) 1, 3 ve 5 mg/l dozları kullanılmıştır. Androgenik performansı yüksek Yamula lokal popülasyonu ile androgenik performansı düşük Anamur F1 genotipleri donör bitki olarak kullanılmıştır. En yüksek ortalama embriyoid oluşumu bal uygulamasında %18.35, şeker uygulamasında %20.95 olmuştur. Yamula daha yüksek androgenik cevap vermiştir. En yüksek embriyoid oluşumu 1,0 mg/l Kinetin + 3,0 mg/l 2,4 D uygulamasından elde edilmiştir. Besin ortamında karbonhidrat kaynağı olarak sakkaroz yerine bal kullanılmasının patlıcanda anter kültüründe embriyoid oluşumu üzerine olumlu etki ederken, besin ortamlarında bal kullanılması ile ilgili daha çok çalışma yapılması gerekliliği ortaya çıkmıştır.
